Researchers Unveil a Promising “Inverse Vaccine” for Autoimmune Diseases

Researchers from the University of Chicago’s Pritzker School of Molecular Engineering have developed a new type of vaccine that can completely reverse autoimmune diseases without shutting down the rest of the immune system. The vaccine removes the immune system’s memory of one molecule, stopping autoimmune reactions like those seen in multiple sclerosis, type I diabetes, or rheumatoid arthritis. Initial phase I safety trials of the vaccine are currently underway.

This “inverse vaccine” marks a significant departure from traditional vaccines, which teach the immune system to recognize and attack viruses or bacteria. Instead, it operates by erasing the immune system’s memory of a specific molecule, thus preventing the body from attacking its own healthy tissues, a phenomenon commonly observed in autoimmune diseases.

The researchers have utilized the liver’s inherent mechanism of marking molecules from deteriorating cells with “do not attack” flags, averting autoimmune reactions to cells that undergo natural death processes. This innovative approach has shown promising results in halting autoimmune reactions associated with diseases resembling multiple sclerosis.

Jeffrey Hubbell, the lead author of the study and the Eugene Bell Professor in Tissue Engineering, emphasized the potential of this approach to treat diseases like multiple sclerosis even after the onset of inflammation, making it a highly practical solution in real-world scenarios.

The study, detailed in Nature Biomedical Engineering, underscores the liver’s role in fostering peripheral immune tolerance, a process that prevents immune reactions to every damaged cell in the body. By attaching molecules to a sugar known as N-acetylgalactosamine (pGal), the team was able to mimic this process, directing the molecules to the liver where tolerance develops.

At present, the treatment of autoimmune diseases involves drugs that broadly suppress the immune system, inadvertently hampering the body’s ability to fight infections and leading to numerous side effects. The inverse vaccine promises a more targeted approach, potentially minimizing side effects significantly.

Phase I safety trials have already been initiated in individuals with celiac disease, with further trials in progress for multiple sclerosis. These trials are being conducted by the pharmaceutical company Anokion SA, a collaborator and funder of this research, alongside the Alper Family Foundation.

As we stand at the threshold of a new era in medical science, the anticipation builds for the advancements this technology promises in the field of autoimmune disease treatment.
